چکیده

Artepillin C (ARC) is a major active ingredient of Brazilian propolis which is a resinous mixed compound collected by honeybeesfrom various plant sources. Previously, we reported that the ethanol extract of Brazilian propolis possesses antiangiogenic activity both in vitro and in vivo. However, the mechanism of angiogenesis inhibitionby purified ARC has not been well-clarified. In this study, we investigated the effects of ARC on tube-forming human umbilical vein endothelial cells (HUVECs). We found that inhibition of tube formation by ARC was accompanied by partial fragmentation of endothelial cells, indicating that it induced cell death. Western blotting revealed that ARC induced the reduction of vascular endothelial cadherinand platelet endothelial cell adhesion molecule-1. ARC also induced chromatin condensation and cell nuclear fragmentation, morphological markers of apoptosis, in tube-forming HUVECs. Furthermore, ARC suppressed phosphorylation of extracellular signal-regulated kinase 1/2, but upregulated phosphorylation of p38. It was also shown that ARC induced apoptosis via activation of proapototic signaling, activation of caspase-3 and cleavage of poly ADP-ribose polymerase and lamin A/C. In conclusion, ARC exerts its antiangiogenic effects through induction of endothelial apoptosis.

Suppression of tumor-induced angiogenesis by Brazilian propolis: major component artepillin C inhibits in vitro tube formation and endothelial cell proliferation.

Propolis, a resinous substance collected by honeybees from various plant sources, possesses various physiological activities such as antitumor effects. We have previously shown that propolis of Brazilian origin was composed mainly of artepillin C and that its constituents were quite different from those of propolis of European origin. In this report, we examined an antiangiogenic effects of Bra...

Myricetin Inhibits Angiogenesis by Inducing Apoptosis and Suppressing PI3K/Akt/mTOR Signaling in Endothelial Cells

Background Myricetin has been shown to possess potential antiangiogenic effects in endothelial cells. However, the underlying mechanisms are not fully understood. Therefore, we evaluated its antiangiogenic effects in human umbilical vascular endothelial cells (HUVECs). Methods HUVECs were cultured in endothelial cell growth medium-2 to induce proliferation and angiogenesis and treated with di...

Brazilian Propolis Suppresses Angiogenesis by Inducing Apoptosis in Tube-Forming Endothelial Cells through Inactivation of Survival Signal ERK1/2

We recently reported that propolis suppresses tumor-induced angiogenesis through tube formation inhibition and apoptosis induction in endothelial cells. However, molecular mechanisms underlying such angiogenesis suppression by propolis have not been fully elucidated. The aim of this study was to investigate the effects of ethanol extract of Brazilian propolis (EEBP) on two major survival signal...

P18 peptide, a functional fragment of pigment epithelial-derived factor, inhibits angiogenesis in hepatocellular carcinoma via modulating VEGF/VEGFR2 signalling pathway

The P18 peptide is a functional fragment of pigment epithelial-derived factor (PEDF), which is an endogenic angiogenesis inhibitor. This study sought to determine the anti-angiogenic bioactivity of the P18 peptide in hepato-cellular carcinoma (HCC) and to elucidate the underlying mechanism. Xenograft tumour growth assays demonstrated the P18 peptide suppressed angiogenesis of HCC in vivo. Wound...

A cassaine diterpene alkaloid, 3β-acetyl-nor-erythrophlamide, suppresses VEGF-induced angiogenesis and tumor growth via inhibiting eNOS activation

Angiogenesis is one of the hallmarks of cancer, playing an essential role in tumor growth, invasion, and metastasis. 3β-Acetyl-nor-erythrophlamide (3-ANE), a cassaine diterpene alkaloid compound from Erythrophleum fordii, exerts various pharmacological effects, including antitumor activity. However, the effects of 3-ANE on tumor angiogenesis and its potential molecular mechanism are still unkno...
